When am I using up my data quota on a iphone 4?

I have unlimited wifi, so i know that whenever i’m connected to wifi, thats ok.
But what if the symbols E and 3G are on the top left of the phone? If i use the internet while those are there, will i be using up some of my data quota?

the GPS is not included on any data plan & is extra to use.
If you are connected to WIFI, it does not count towards your data usage regardless of whether you have 3G or EDGE.
